704w\" sizes=\"(max-width: 606px) 100vw, 606px\" \/><\/figure>\n\n\n\n<p>In this new\u00a0<em><strong>Science<\/strong><\/em>\u00a0paper, our collaborators in the Baker lab used AI-guided design to create proteins that bind tightly and specifically to 39 disordered targets, including the opioid peptide dynorphin, cancer-related receptor regions, and prion-like viral sequences. Together with X-ray crystallography, <strong>our lab validated <\/strong>the dynorphin binders by <strong>NMR<\/strong>, confirming that binding locks the flexible peptide into a defined shape. This modular method can be used to <strong>design binding proteins for virtually any disordered protein or peptide target allowing for the ability to now target the &#8220;untargetable&#8221;.<\/strong><\/p>\n\n\n\n<p class=\"has-text-color has-link-color wp-elements-7a9d00d87b19c9e5d2962be695918449\" style=\"color:#7fa9ae\"><strong><a href=\"https:\/\/www.nature.com\/articles\/s41598-024-81990-y\">An isothermal calorimetry assay for determining steady state kinetic and Ensitrelvir inhibition parameters for SARS-CoV-2 3CL-protease<\/a><\/strong><\/p>\n\n\n\n<p>This manuscript details the application of Isothermal Titration Calorimetry (ITC) to characterize the kinetics of 3CL<sup>pro<\/sup>, the main protease from the Severe Acute Respiratory Syndrome CoronaVirus-2 (SARS-CoV-2), and its inhibition by Ensitrelvir, a known non-covalent inhibitor. 3CL<sup>pro<\/sup>&nbsp;is essential for producing the proteins necessary for viral infection, which led to the COVID-19 pandemic. The ITC-based assay provided rapid and reliable measurements of 3CL<sup>pro<\/sup>&nbsp;activity, allowing for the direct derivation of the kinetic enzymatic constants&nbsp;<em>K<sub>M<\/sub><\/em>&nbsp;and&nbsp;<em>k<sub>cat<\/sub><\/em>&nbsp;by monitoring the thermal power required to maintain a constant temperature as the substrate is consumed.<\/p>\n\n\n\n<p class=\"has-text-color has-link-color wp-elements-05494f8efdb2f8d9c47a665a54ff9f59\" style=\"color:#7fa9ae\"><\/p>\n<\/div>\n\n\n\n<div class=\"wp-block-column
